B型肝炎病毒(hepatitis B virus,HBV)感染是導致肝臟病變的隱形殺手,長久以來危及人們的健康。B型肝炎病毒基因型分為(A~H)8型,不同基因型的病毒,會導致病毒在基因、結構、以及臨床表現上的差異,此外病毒序列的變異對感染者於病程趨向之影響,亦是B型肝炎相關研究者積極探討之問題。因此本研究將應用資料探勘(Data Mining)技術,由病毒胺基酸序列的表現來辨別出(A~H)8種基因型,並且指出與臨床因子(如:基因型、病毒量高低、血清轉換與否等…)相關之胺基酸序列位置,此外,並設計一統計分析系統,計算大量病毒序列的突變率以及根據各種不同分組條件來進行統計分析。研究結果顯示由P基因第73位置、104位置以及199位置,即可將NCBI資料庫358株病毒序列,分辨出 (A~H)8個基因型,其準確率達100%。並且由病人抽血第一時間點的病毒胺基酸序列,歸納出可判定病人是否有走入血清轉換的或已達血清轉換的趨勢之準則。也根據不同時間點病毒的突變率,指出病毒在病程中突變演化的動力假說。
The hepatitis B virus infection is the invisible killer of pathological changes of liver, and endangered people''s health for a long time. The hepatitis virus genotype has 8 different types (A to H), and genotypes have different genes, and structures, and induces different results. In addition, the influence of the variation of the virus sequence is also our study in this thesis.This research will apply data mining technique to distinguish the genotypes from the amino acids sequence of the virus, and point out the relation between amino acids sequence position and the clinical results. In addition, we designed a statistical analysis software for calculating mutation rate from the mass virus sequences and did some statistical analyses on some conditions predefined by users. Analysis results show that we can distinguish 8 genotypes according to the 73th, 104th and 199th positions of the P gene among 358 virus sequences in the NCBI database, and its correct rate reached 100%. We can also predict the HBeAg of patients to be seroconversion or not. According to amino acids sequence of virus in the first sampled blood, we can predict the trend of HBeAg seroconversion. Finally, we leads the motive hypothesis that the evolution of the virus in disease exits the high relationship with the mutation rate in different time periods.
